It is a metabolite of another synthetic analog known as melanotan II. Early research indicated that melanocortin hormones can influence various physiological functions. When test animals were given the isolated hormone, it was observed to increase reproductive activity, which remains one of the main areas of investigation for PT-141 peptide.

Overview Studies suggest that PT-141 exhibits agonistic effects on melanocortin receptors, particularly MC3R and MC4R, which may lead to heightened activity in the central nervous system. MC3R is mainly expressed in the brain, especially the hypothalamus, where it may contribute to energy balance. Research groups studying melanocortin receptors have proposed that MC3R could help regulate the activity of other melanocortin receptors in this region. It may also affect feeding behavior, appetite, and metabolic processes involving glucose and lipids.(3) MC4R, on the other hand, is thought to play a key role in appetite control. When activated in the brain, it is believed to help regulate food intake. MC4R is also linked to energy expenditure, with activation potentially increasing energy use and supporting weight management. This receptor has additionally been associated with reproductive function, including effects on penile tissue and overall reproductive behavior.(4) In published studies, researchers noted that binding to MC3R and MC4R receptors appeared to activate neurons in the hypothalamus, resulting in increased immunoreactivity.
